ConviFlex™ RT-Taq Mix is a lyophilized, one-step RT-PCR master mix, consisting of all core components required for reverse transcription and PCR amplification. Only user-based RNA templates, primers, and probes (for RT-qPCR) are required to carry-out in one step a complete RT-qPCR-based analysis.
RT-qPCR is an indispensable tool in molecular biology to perform both quantitative and qualitative analysis of RNA molecules. Reverse transcription of purified RNA and subsequent PCR amplification of cDNA are usually performed sequentially, in several pipetting and incubation steps.
Using ConviFlex™ RT-Taq Mix for several RT-PCR-based applications requires only few pipetting steps and minimal hands-on time, which reduce the margin of error, the risk of cross-contaminations and work time.
In ConviFlex™ RT-Taq Mix, a MuLV reverse transcriptase, a hot-start Taq polymerase, and dNTPs are pre-mixed and lyophilized for convenient, efficient and reproducible RT-PCR. The reverse transcriptase included in the mix is genetically modified from murine leukemia virus (M-MuLV). The Taq polymerase included in ConviFlex™ RT-Taq Mix has a hot start functionality: its activity is blocked at temperatures lower than 70 °C through anti-Taq DNA polymerase antibodies. This feature strongly suppresses the formation of unspecific PCR products and primer-dimers, resulting in PCR reactions with higher specificity, increased sensitivity, and greater yields.
Once resuspended in the supplied Rehydration Buffer, the master mix contains optimal concentrations of Mg2+, deoxyribonucleotide triphosphates, and stabilizers for excellent RT-qPCR performance.
ConviFlex™ RT-Taq Mix is a universal master mix, which can easily be used in most RT-PCR protocols, programs and cycler types. Upon addition of gene-specific primers and target RNA, the user can start the RT-PCR reaction without intermediate steps between the reverse transcription and the PCR.
Intended for research use only. Not recommended for clinical diagnostics. ConviFlex™ RT-Taq Mix is designed for convenient detection and analysis of RNA molecules by one-step RT-PCR and is compatible with both conventional RT-PCR and RT-qPCR. RNA templates may be extracted from diverse starting materials using a commercially-available kit (see ExtractNow™ Virus RNA Cat. No. 611-1250) or a well-established extraction method. Optimal volumes, concentrations, temperatures and incubation time may greatly vary for your specific application and should be adjusted to the requirements of your own assay.
