Convenient Mycoplasma Detection

 

Venor®GeM Advance is a ready-to-go version of Venor®GeM. The reaction tubes included with the kit are pre-coated with appropriate dNTPs and primers to reduce the total assay time compared to the general protocol that requires individual loading of reaction tubes. For additional convenience the gel loading buffer and dye are already included in the reaction tubes. After thermal cycling the PCR can be loaded directly on the agarose gel.

Benefits of Venor®GeM Advance

  • Sensitive
    Detection of 5 copies/µl of mycoplasma DNA that corresponds to 10 mycoplasma genomes per sample volume.
  • Specific
    The kit is specific for more than 79 mycoplasma species including all relevant species mentioned in the Eur. Ph. 2.6.7. For the detection of the avian associated mycoplasma species M. synoviae and M. gallisepticum the product Venor®MGS is highly recommended. Eukaryotic and bacterial DNA is not amplified.
  • Reliable
    The tubes contain internal control DNA for individual inhibition control. The pre-aliquoted reagents optimize reproducibility and minimize operator-dependent variability of results.
  • Simple
    Only rehydration buffer, polymerase and sample have to be added to the PCR tube.
  • Clear
    A clear and easily interpretable result is obtained with one PCR reaction.

Test Principle

Venor®GeM Advance utilizes the polymerase chain reaction, which is established as the method of choice for highest sensitivity in the detection of mycoplasma and acholeplasma contamination in cell cultures and other cell culture derived biologicals. The primer set is specific to the highly conserved 16S rRNA coding region in the mycoplasma genome. This allows for detection of all mycoplasma species usually encountered as contaminants in cell cultures. The reaction tubes also contain DNA to serve as an internal control. For the internal control DNA, a successfully performed reaction is indicated by a distinct 190 bp band on the agarose gel. A separate strip of vials contains non-infectious Mycoplasma orale DNA as a positive control.

 

Background

Mycoplasma are known to cause a variety of human and animal diseases, and in the laboratory, mycoplasma are often found as common contaminants in cell cultures, virus stocks, and other cell-derived biologicals. Mycoplasma contamination is a recurring problem that can cause non-reproducible results in research and productivity losses of up to 50% in industry, which can be avoided by routine diagnostic testing. Also, testing for mycoplasma infection is now an essential requirement in reaching quality control standards for laboratories concerned with the production and maintenance of cell lines used in manufacturing various biologicals.

We highly recommend our reliable hot-start MB Taq DNA Polymerase (Cat.-No. 53-0200) with Venor®GeM Advance. Use of other Taq polymerase products may require reaction optimization to achieve proper results.

Venor®GeM Advance kit components are lyophilized and at +2 to +8 °C maintainable for at least 12 months. Primers and nucleotides are aliquoted in individual PCR reaction tubes to guarantee consistent quality. After rehydratisation the reagents must be used within 1 hour.

 

 

Venor®GeM Advance is intended for research use only. Not recommended for clinical diagnostics or testing of human samples.
