MB Taq DNA Polymerase

Hot-start MB Taq DNA Polymerase is the optimized mixture of Taq (Thermus aquaticus) DNA Polymerase and anti- Taq DNA polymerase monoclonal antibodies. Antibodies block polymerase activity during set-up of the PCR reactions at ambient temperature (20-22°C). The inhibition of Taq DNA Polymerase is completely reversed when the temperature is above 70°C. This enzyme is optimal for all Minerva Biolabs products except Onar®EUB. Complete activation of the polymerase is achieved at 94 °C for 2 min.


“Hot Start” Feature

The “hot start” Taq mixtures are designed to prevent unwanted amplification by preventing polymerase activity until the reaction mixture is above a certain temperature (usually accomplished using a monoclonal antibody directed against Taq).

Application

Complex genomic or cDNA templates, low copy number targets, large numbers of thermal cycles, multiplex PCR.



Unit Definition

One unit is defined as the amount of the enzyme required to incorporate 10 nmoles of dNTPs into acid-insoluble DNA fraction in 30 minutes at 72°C.



Contents and Storage

MB Taq DNA Polymerase, supplied at 4000-5000 units/ml, 4-5 units/µl in 10 mM Tris-HCl (pH 7.0), 50 mM KCl, 0.1 mM EDTA, and 50% (v/v) glycerol. 10x reaction buffer and 100 mM MgCl2 -solution. Store below -18°C.
