PCR Analysis of DNA Extract
Our detection services for Legionella spp. and L. pneumophila of already prepared DNA extracts allows customers the convenience of collecting and preparing water samples on-site to conform with individual testing guidelines. The PCR analysis can then be outsourced to our expert testing laboratory. The Aqua Screen® PCR system is a quantitative test for the detection of Legionella species and/or L. pneumophila in water samples. Detailed validation studies confirm the high specificity and sensitivity of Aqua Screen®. Clinically relevant species detected by the system include L. pneumophila SG 1-14 and SG 16, L. longbeachae, L. dumoffii, L. bozemanii, L. gormanii, L. anisa, L. cherrii, L. parisiensis, L. tucsonensis, L. wadsworthii.
Cross activity with other species typical for water contamination is not known. The detection limit is 60 genome equivalents per 100 ml sample volume (corresponding to 1 CFU through the ISO 11731-2 method) following filtration of 1 l.
Sample Preparation and Shipment
As the validity of the test results are subject to the sampling conditions, we highly recommend the use of the Aqua Screen® Filtration Unit (31-0300) and Extraction Kit (32-0150) to ensure optimal results. The DNA extract volume should be 100 µl. The DNA extract can be dispatched at ambient temperatures following heat treatment (10 min. at 95 °C). Samples are accepted only with this completed and signed Submission Form.
Confidentiality/Quality Assurance
All client information is treated in strict confidence. Formal, contractual confidentiality agreements can be generated upon clients' request. Reports are maintained in our archives for a minimum of five years. Extracted samples are kept at Minerva Biolabs for up to six months for retesting at the customers' request. The laboratory practices GMP and GLP standards and is ISO 13485 and 9001:2000 certified.
