Benefits of Venor®GeM-qEP

  • Approved
Validated according to the European Pharmacopoeia 2.6.7. for the testing of cell culture supernatants of master and production cells, chondrocytes for human application, sera, cell culture media, and therapeutic antibody formulations.
  • Sensitive
Individual primer probe sets maximize the sensitivity for Mycoplasma species, A. laidlawii and M. pneumoniae.
  • Specific
Validated for mycoplasma species listed in the European Pharmacopoeia:
    A. laidlawii, M. fermentans, M. hyorhinis, M. orale, M. pneumoniae, M. synoviae, M. argininii, Spiroplasma citri (add-on required). Detects more than 67 mycoplasma species. No detection of Arthrobacter, Burkholderia and other water born DNA contaminants.
  • Simple
Robust method, no need for general DNA extraction with cell culture supernatants.
  • Reliable
Kit contains internal control and positive control DNA and a specific internal control probe for optional PCR quality check.

Test Principle

Venor®GeM-qEP can be used for the detection of mycoplasma DNA in biological samples, like cell cultures. The specific primers amplify nucleotide sequences within a highly conserved region of the 16S rRNA gene specific for Mycoplasma species, M. pneumoniae and Acholeplasma laidlawii in individual PCR reactions which can be processed in the same cycler run. To rule out the presence of PCR inhibitors in the sample, an internal control is included in Venor®GeM-qEP. Venor®GeM-qEP can also be used with UNG for carry-over prevention. The optional Quantification Standards (page 66) provide titrated DNA for the quantification of the mycoplasma DNA load in the sample.

Background

Mycoplasma are known to cause a variety of human and animal diseases, and in the laboratory, mycoplasma are often found as common contaminants in cell cultures, virus stocks, and other cell-derived biologicals. Mycoplasma contamination is a recurring problem that can cause non-reproducible results in research and productivity losses of up to 50% in industry, which can be avoided by routine diagnostic testing. Also, testing for mycoplasma infection is now an essential requirement in reaching quality control standards for laboratories concerned with the production and maintenance of cell lines used in manufacturing various biologicals.

Kit Components

  • primers, probes and nucleotides at optimized concentration
  • reaction buffer optimized for MB Taq DNA Polymerase
  • positive control for easy result verification
  • internal control to ensure accurate testing
  • PCR Grade Water for resolving the components* and setting up the mastermixes

We highly recommend our reliable hot-start MB Taq DNA Polymerase for PCR detection of mycoplasma (Cat. No. 53-0200).

Venor®GeM-qEP kit commponents are lyophilized and at +2 to +8 °C maintainable for at least 12 months. Primer and nucleotides are aliquoted
(á 25 tests) to guarantee consistent quality. After rehydratisation the reagents must be stored below -18 °C.
