Background

Treponema pallidum is causing Syphilis, also called “Lues”. Syphilis is a curable sexually transmitted disease caused by the Treponema pallidum spirochaete. The organism can also be transmitted to a fetus by transplacental passage during the later stages of pregnancy. Syphilis can be easily treated with antibiotics including penicillin. If not treated, syphilis can cause serious effects such as damage to the heart, aorta, brain, eyes and bones.

Test Principle

The Onar®Tp test system is a test for a qualitative and quantitative diagnostics of in clinical samples. The test is based on the polymerase chain reaction and allows for rapid diagnostics of infections. The supplied primer, probes and the positive control are specific for a segment of the polA gene of the Treponema pallidum genome. When using the Onar®Tp Detection Kit for real-time PCR the specific target probe is emitting a fluorescent signal at 520 nm. The realtime PCR internal control is detected by another probe emitting fluorescent light at 600 nm. For sampling and sample transport sterile swabs without transport medium should be used (100% Darcon® or Nylon Swabs). The material can be directly analyzed from the swab. Swabs of ulcer and vaginal smear can be use as test material.

Typical results by performing the Onar®Tp PCR on the LightCycler. 105– 101 genome copies of Tp have been taken as template. The specific fluorescent signal was detected in channel F1.
