The Onar®MRSA is an in vitro test for quantitative and rapid PCR-based detection of Methicillin-resistant Staphylococcus aureus in clinical samples. This one-step triplex test system is a CE-marked IVD and approved for in vitro clinical diagnostics. The test system has undergone extensive validation programs.

Background

MRSA are specific strains of Staphylococcus aureus which have developed antibiotic resistance to all penicillins, including methicillin and narrow-spectrum ß-lactamase-resistant penicillin antibiotics. MRSA can infect other tissues when normal barriers have been breached (e.g. skin). This leads to furuncles, heavy infections and can lead to a sepsis. The Methicillin-resistant Staphylococcus aureus is described as a trigger for typical hospital related infections.

Methicillin-resistant Staphylococcus aureus are diagnosed by amplification of the mecA gene located within the Sccmec gene locus. The subsequent discrimination of the human pathogenic Staphylococcus aureus and the commensally present Staphylococcus epidermidis will be performed by utilizing the femA gene for amplification.

 

Test Principle

The Onar®MRSA is a test for semi-quantitative diagnostics of clinical samples. The test can basically be used with all relevant PCR machines. The supplied primer, probes and the positive control are
specific for a segment of the mecA gene of the Methicillin- resistant Staphylococcus aureus genome and the Staphylococcus aureus verification gene femA. The target probe emits fluorescent light at
~ 520 nm and the S. aureus confirmation emits fluorescent light at
~ 610nm. To rule out the presence of PCR inhibitors in the individual patient specimens, an internal control is included in the kit. The internal control is detected by another probe emitting light at ~ 560 nm.
