Chlamydia trachomatis for qPCR

 

The Onar®Ct is an in vitro test for quantitative and rapid PCR-based detection of Chlamydia trachomatis (Ct) DNA in various clinical samples. These samples should be extracted from infected or routine-screening patients. Patient specimens like urine, pool-urine and ejaculates as well as urogenital swabs are used as starting material for the DNA Extraction.

 

The Onar®Ct test system is CE-marked according to the European IVD Directive 98/79/EC.

    Onar®Ct is designed to amplify the chromosomal gene momp (major outer-membrane protein). The momp gene is a single copy gene which is suitable for quantification. Furthermore the test detects the new variant Chlamydia trachomatis (nvCT) representing a deletion in the cryptic plasmid, (MØller JK et al., 2008, Alexander S. and Ison C., 2008).

    The kit contains a positive control DNA and a specific internal control probe for optional PCR quality check. Results are obtained within 2-3 hrs.

    sample preparation methods including urine pools allow an instant and simple application of the kit.

Background

Chlamydia trachomatis is an intracellular, gram-negative bacterium. Infection of the lower genital tract is one of the most prevalent sexually transmitted diseases (STD) in the world. Chlamydia trachomatis infection may cause urethritis, cervicitis and pelvic inflammatory disease (PID) in woman. Detecting chlamydial genital infection and preventing transmission and spread of this infection to the upper reproductive tract are a challenge for both clinician and laboratory workers. Nucleotid acid amplification techniques are more used to diagnose chlamydial infections.

 

Test Principle

Onar®Ct is a test for quantitative diagnostics of clinical samples. The test can basically be used with all relevant PCR machines. The supplied primer, probe and the positive control are specific for a segment of the momp gene of the Chlamydia trachomatis genome. The target probe emits fluorescent light at ~520 nm. To rule out the presence of PCR inhibitors in the individual patient specimens, an internal control is included in the kit. The internal control is detected by another probe emitting light at ~610 nm (Type 1) or ~560 nm (Type 2).
